Yū Amagi is a slightly eccentric detective who has been assigned to the Twelfth Section of the Tokyo Metropolitan Police Department's First Investigative Division from the Lost and Found Centre. The Twelfth Section is called the "graveyard of detectives" and ridiculed as the "banishment room where hardened detectives who cannot be fired are sent." Its assembled detectives Kōsuke Samura, Takumi Yamashita, Keita Nagasawa, Tamaki Mizuta, and Masatoshi Katagiri are elite and yet oddballs. Amagi appears fixated with time which seems to have no relation with a case. Raising questions about the timelines of the perpetrator and victim derived from the estimated time of death, time of crime, time of alibi, and time limit, he searches for the meaning of "blank time" which arises from this. He obsesses over the weight of every minute and second of time because of some incident.

Yukiko Makabe works at the #3 Special Investigation Team of the Metropolitan Police Department. She has excellent negotiation skills. Yukiko then attempts to negotiate with a hijacker on a bus, but something unexpected happens and her mission fails. Yukiko takes full responsibility and takes a transfer to a special interrogation team "Kintori". Her boss is Katsutoshi Kajiyama and Yukiko has had a difficult working relationship with him in the past. Her other coworkers include Susumu Hishimoto, Zenjiro Nakata and Haruo Koishikawa.

Enomoto Kei is a security "otaku" working for a security firm, devoted to improving the securities system on a daily basis. He is not an easy person to familiarize oneself with, always collected and calm, unapproachable, a maniac in Physics, Science, Architecture and profound in other basic theoretical foundation. He works side by side with Aoto Junko and Serizawa Gou, who are lawyers working at a major law firm. Junko is pure and forward, acting upon her instincts while Serizawa is a prideful elitist who sees time as money, and therefore, will not take on any job that is unprofitable. Possessing a vast amount of knowledge and acute insightfulness, Kei is able to unlock the closed doors and unravel the secrets leading to the solution. Never before has there been a drama focused solely on closed doors.

Satoshi Itomura is an assistant police investigator who belongs to the Metropolitan Police Department’s Scientific Investigation Team. Reenactments, fingerprint and voice data collections, profiling... His team collects scientific evidence and pieces together fragments of information in order to help build evidences. However, Itomura often does not care about the scientific approach and instead pays a great deal of attention to the things that the victims always carried with them or treasured. And he focuses on these things because they usually carry the victim’s dying wishes, understanding that last message not only helps solve the cases but also brings closure to the depressed family of victims.

Mizuki plays a 35-year-old woman named Kurumi, who suddenly loses everything in her life when her fiance runs off with all of her assets. Left with a mere 15,000 yen and some boxes of ohagi (a type of rice ball), the despairing Kurumi happens upon a construction site, and she begins handing out the ohagi to the men working there. Using the theme of food, "Tenshi no Wakemae" depicts Kurumi's journey as she turns her life around.
